To provide comprehensive molecular biology technical support for postdoctoral fellows to investigate structure and function of ion channels. Required techniques include: preparation of competent cells and Gibson assembly reagents; primer design, PCR and all stages of cloning for making constructs in traditional and CRISPR-based vectors; analysis of DNA sequences; mini and large preparation of plasmid DNA; preparation of total and mRNA for Northern blot analysis and cDNA library construction; preparation of genomic DNA from fly and mouse tissues; generation of Bacmid and Baculovirus for protein expression. To establish primary cell culture and maintain human tumor cell lines; to perform subcutaneous injection of tumor cells into mice and rats to establish tumor model; to perform protein quality analysis by SDS-PAGE, Western blot analysis and co-Immunoprecipitation assay; to preserve all published constructs from the lab; to sort, aliquot, store and monitor lab antibody stocks..
